Your Own Site's RSS Feed
This may not seem like an obvious choice, but since it can be used not only with Twitter, but in the RSS feed readers of many a viewer, you will need to treat it as a tool in your arsenal.
Myspace is in a unique position, because it allows for both having a network of friends and public viewing. Unlike a Facebook page, this ability to be viewed by the public makes the links on this page particularly valuable. It also allows people who are not willing to commit to having you as a friend to see what you've posted.
Facebook also has its own unique advantages, since every user who is getting your updates has chosen to receive them. This means that you can be a bit more familiar with your users. That is not to say that you can be lax, or use profanity, but adding cool insider photos or contests will work well here.
Either used alone or with your RSS feed, Twitter is a great way to reach a large number of people with a very timely message. Just do not expect things to linger, as the lives of messages in the Twitter world are measured in seconds.

Tips on Building a Profile
Be honest. Pretending that you are in a demographic that you are not is not only wrong, but you will be seen through in a heartbeat by actual members of it.
Fill out the profile interest section in a well-rounded way. If your interest is in promoting a gaming site, your interests should include specific consoles, genres and titles.
Don't forget images. You may be there to work, but others are there to have fun. Appeal to their asthetics.
Now that you have created a sound profile, all you will have to do is get some friends. So who should you look to for your first set of friends? Well, you might try out:
Any employees you may have.
Any contractors or vendors with which you currently have a relationship.
Any clients with which you have (or have had) good relations and no ill will.
On Twitter, you can try following people with similar interests to gain follow backs from relevant users. Just be aware that this tactic has varying success rates.
